It definitely helps to know how to throw from both sides, because there may come a time when using a throw is appropriate but there are obstacles that prevent you from throwing from your comfortable side. The way you've worded your question is a bit confusing to me, though, because it doesn't matter what hand is forward when you throw, it only matters where you grab them and what direction you turn.

That question confused me too, but in a way that's familiar to me - it's a similar confusion as i'm sure a wrestler/judoka feels when someone says "Well, what if they don't want you to grab them?"

I am not ambidextrous, and I do a symmetrical art. You just learn how to do it is all, same as every other skill.

Being ambidextrious won't be necessary; just make sure to practice executing on both sides. Proficiency will come with time and practice.
